Phenyl-type columns provide strong pi-pi interaction and offer selectivity complementary to other reversed-phase columns (e.g., C18). These columns have been successfully used to separate positional isomers, tocopherols, flavonoids, polynuclear aromatics and nitroaromatic compounds, active pharmaceutical ingredients (APIs) and related compounds. Early phenyl columns contained short alkyl phenyl ligands covalently bound to the silica surface.

    UHPLCS Phenyl HPLC Column is a matrix of super pure silica gel, and it is bonded to propyl phenyl on the surface of silica gel, and is processed with a double–end bonding process. The column can be used for the separation of polar and aromatic compounds.

    1. When should phenyl columns and cyano columns be considered?

    - When the phenyl column is used for the determination of aromatic compounds containing benzene rings, it has higher selectivity. The CN column can be used as a reversed-phase column with the weakest retention capacity, or as a normal-phase column with reduced activity (retention times are much lower than silica and amino columns).

     

    2. When testing samples, I often worry about whether the column pressure of the chromatographic column is too high, but when purchasing the HPLC column, it does not say what is the maximum operating pressure of each HPLC column. In this case, how can the user judge whether the column pressure is too high? Exceeded the limit of the bar?

    - The pressure when the chromatographic column is installed will be much higher than when it is used, so there is no limit to the column pressure when using the chromatographic column itself, but there is an upper limit of 40Mpa for general instruments. If the chromatographic resolution can still meet the requirements of the analytical method, the column pressure is OK as long as the instrument can bear it.

     

    3. I ran into a problem like this before. The chromatographic conditions are mature and the retained substances operated under absolutely no problem conditions are not retained. For example, the retention time is 15 minutes. No matter how the ratio is adjusted, the peak will appear in 2 to 3 minutes, but after maintenance, it will be normal again next time, what is the reason?

    - Most likely a phase collapse occurred. The C18 column and the high-density bonded and sealed C8 column will collapse under the high aqueous mobile phase, and the consequence is that the retention capacity is greatly reduced. After flushing with a mobile phase with a higher proportion of organic phase, the normal performance can be restored.laboratory illustration
