The ÄKTA oligopilot plus is a flexible, fully automated DNA/RNA synthesizer designed for use in R&D and process development laboratories and oligonucleotide production plants.

 

The structure of the DNA synthesizer contains the synthesis column, the flow throttle, and other components.

DNA Synthesis Columns:

Starting from a disposable column containing nucleotides bound to a carrier (usually CPG), not only the column but also 2 fixed hplc column frits and 2 connectors, made of inert material with all components.

 

The hplc column joints at both ends of the column are equipped with hplc frits, which are sintered stainless steel with pore sizes ranging from 5 to 10 μm, depending on the packing size, to prevent packing leakage; at the same time, the sieve plates also have the function of distributing fluid. It has been shown that the thickness and relative diameter of the sieve plate and the aperture diameter can affect column efficiency. Therefore, the column efficiency design based on frit can be improved.

By optimizing the design and ratio of the sieve plate at the inlet of the column and combining it with the diffusion of the mobile phase in the column, the difference in the flow velocity in different radial directions in the column is reduced, the radial asymmetry of the fluid flow pattern in the column is weakened, the asymmetry of the peaks is effectively reduced, and the number of theoretical plates is significantly increased, which effectively improves the column efficiency.
